Membrane Curvature: How BAR Domains Bend Bilayers
نویسندگان
چکیده
An important new structure suggests the BAR domain is a membrane-binding module that can both produce and sense membrane curvature. BAR resembles a banana that binds membranes electrostatically through its positively charged, concave surface.

Structural Basis of Membrane Invagination by F-BAR Domains
BAR superfamily domains shape membranes through poorly understood mechanisms. We solved structures of F-BAR modules bound to flat and curved bilayers using electron (cryo)microscopy. We show that membrane tubules form when F-BARs polymerize into helical coats that are held together by lateral and tip-to-tip interactions. On gel-state membranes or after mutation of residues along the lateral int...
متن کاملStructural Basis of Membrane Bending by the N-BAR Protein Endophilin
Functioning as key players in cellular regulation of membrane curvature, BAR domain proteins bend bilayers and recruit interaction partners through poorly understood mechanisms. Using electron cryomicroscopy, we present reconstructions of full-length endophilin and its N-terminal N-BAR domain in their membrane-bound state. Endophilin lattices expose large areas of membrane surface and are held ...
متن کاملModeling membrane deformations and lipid demixing upon protein-membrane interaction: the BAR dimer adsorption.
We use a self-consistent mean-field theory, designed to investigate membrane reshaping and lipid demixing upon interaction with proteins, to explore BAR domains interacting with large patches of lipid membranes of heterogeneous compositions. The computational model includes contributions to the system free energy from electrostatic interactions and elastic energies of the membrane, as well as s...
متن کاملMembrane stiffness is modified by integral membrane proteins
The ease with which a cell membrane can bend and deform is important for a wide range of biological functions. Peripheral proteins that induce curvature in membranes (e.g. BAR domains) have been studied for a number of years. Little is known, however, about the effect of integral membrane proteins on the stiffness of a membrane (characterised by the bending rigidity, Kc). We demonstrate by comp...
متن کاملBin/Amphiphysin/Rvs (BAR) family members bend membranes in cells
We provide direct evidence that Bin/Amphiphysin/Rvs (BAR) family members bend the steady state membrane architecture of organelles in intact cells. In response to inducible BAR molecular actuators, organelles exhibit distinct changes to the orientation and degree of their membrane curvature. This rapidly inducible system may offer a mechanism by which to better understand the structure-function...
